I think it depends on how heavy a user you are, and what it is you're likely to tweet.
Personally, I find the website is fine because I only follow a few people and I tweet rarely. On my phone I use the app it came with. My friends who follow a lot of people use Tweetdeck.
I use my real name because I don't tweet anything controversial, or frankly anything interesting. I'm @youRwired which is also my domain name so people can remember it. Most of my friends use their on-line alias. I was going to use my forum name, but annoyingly it's already taken.

If I want to be able to tweet from multiple accounts e.g. work and personal, does that influence the app decision?
I suspect my phone will be used far more than a computer. Especially overseas.

Third party Twitter apps are being restricted in what they can do, connections they can make, size of user base, so if I was choosing now, I'd prolly try the official Twitter app first. I'm @SFGPhoto.

Like Mark, I use the official Twitter app on my Mac. It's no longer being supported but while it ain't broke I won't fix it. I don't feel any need to buy an app that does way more than I need.

I use tweetdeck as I run multiple accounts (but haven't found a client I'm willing to pay for yet).
I have 2 main accounts.
One in my "online name" (i.e. saspro) and another that's my real name.
That way I can separate out business contacts to their own account.

Would you say Tweetdeck is preferable if you potentially want to run two accounts then Sas? Or can that still be done with Twitter? Could I install both on the phone to try them, or is that asking for trouble?

I use the twitter app an my phone with 2 accounts and it works fine. On the mac I have to use an app as unless I use multiple browsers I can't do what I need (as I actually use 12 accounts)

I'm still on the old Tweetdeck (Air based) on my iMac and Windows PC!
